PTEN Colorimetric Cell-Based ELISA Kit

Assay Genie offers this Cell-Based ELISA Kit for the detection of PTEN (Phosphatase and Tensin Homolog) protein in cell lysates and tissue homogenates. The assay is intended for adherent cells and uses a colorimetric detection method at 450 nm.

The kit shows reactivity with Human, Mouse, and Rat samples, with a dynamic range of > 5000 Cells. Storage stability is specified at 4°C for 6 months. The product is designed as a convenient, lysate-free, high-throughput and sensitive assay that can be used for measuring the relative amounts of PTEN in cultured cells, as well as screening for the effects that various treatments, inhibitors (such as siRNA or chemicals), or activators have on PTEN.

Qualitative determination of PTEN concentration is achieved by an indirect ELISA format: PTEN is captured by PTEN-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control in normalising the target absorbance values. Following the colorimetric measurement, a Crystal Violet whole-cell staining method may be used to determine cell density, allowing results to be analysed by normalising absorbance values to cell amounts and adjusting for plating differences.
