Description

Assay Genie offers a Cell-Based ELISA Kit designed for the quantitative measurement of PSMD2 levels in cell lysates and tissue homogenates. The kit targets PSMD2, also known as 26S proteasome non-ATPase regulatory subunit 2, a regulatory subunit of the 26S proteasome that participates in the ATP-dependent degradation of ubiquitinated proteins. Dysregulation of PSMD2 has been implicated in various diseases, including cancer and neurodegenerative disorders, highlighting its importance as a potential therapeutic target.

This lysate-free, high-throughput assay uses an indirect ELISA format. PSMD2 is captured by PSMD2-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. The conjugated HRP enzyme catalyses a colorimetric reaction upon substrate addition, measured at 450 nm. The kit is reactive with Human, Mouse, and Rat samples, with a dynamic range of > 5000 cells, and is provided in a 96-well microplate format. It can be used to measure relative amounts of PSMD2 in cultured cells and to screen for the effects that various treatments, inhibitors (such as siRNA or chemicals), or activators have on PSMD2.

A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing the target absorbance values. Following colorimetric HRP activity measurement, the Crystal Violet whole-cell staining method can be used to determine cell density, enabling normalization of absorbance values to cell amounts to adjust for plating differences. The kit is stored at 4°C with a stability of 6 months. Database identifiers for the target include Gene ID 5708, UniProt ID Q13200, OMIM 606223, and Unigene Hs.518464.
