Description

A colorimetric cell-based ELISA kit from Assay Genie designed for the qualitative detection of phosphorylated PSEN1 (Presenilin-1) at Ser357 in cultured adherent cells. The assay uses an indirect ELISA format in which a phospho-specific primary antibody captures PSEN1 (Phospho-Ser357) and an HRP-conjugated secondary antibody catalyses a colorimetric reaction read at 450 nm upon substrate addition.

The kit is lysate-free and supports high-throughput screening of the effects of various treatments, inhibitors (e.g. siRNA or chemicals), or activators on PSEN1 phosphorylation. It is reactive with Human, Mouse, and Rat samples, with a dynamic range of > 5000 Cells, and is supplied in a 96-well microplate format. Normalisation is achieved using an included anti-GAPDH monoclonal antibody as an internal positive control together with a Crystal Violet whole-cell staining step to determine cell density and adjust for plating differences.

PSEN1 is a probable catalytic subunit of the gamma-secretase complex, an endoprotease complex that catalyses the intramembrane cleavage of integral membrane proteins such as Notch receptors and APP (beta-amyloid precursor protein), and plays a role in the generation of beta-amyloid peptides implicated in neurodegenerative disorders including Alzheimer's disease. Phosphorylation at Ser357 is reported to play a crucial role in PSEN1 function, with dysregulation associated with neurodegenerative disease, making accurate measurement of this modification in cell lysates useful for research into disease mechanisms and potential therapeutic targets.
