Proteasome alpha3 Colorimetric Cell-Based ELISA Kit

A cell-based ELISA solution from Assay Genie for the qualitative measurement of proteasome alpha3 protein expression in cultured cells. The kit uses an indirect ELISA format, in which target-specific primary antibodies capture proteasome alpha3 and HRP-conjugated secondary antibodies bind the Fc region of those primaries; the conjugated HRP enzyme then catalyses a colorimetric reaction upon substrate addition, measured at 450 nm.

The assay is reactive with human, mouse and rat samples and is suitable for adherent cells, enabling the relative quantification of proteasome alpha3 expression as well as screening of treatment effects, inhibitors such as siRNA or chemicals, or activators. A dynamic range of greater than 5,000 cells is supported. For data normalisation, a monoclonal antibody specific for human GAPDH is included as an internal positive control, and a Crystal Violet whole-cell staining step allows absorbance values to be normalised to cell density, correcting for plating differences.

Proteasome alpha3 (gene symbol PSMC3, UniProt ID P17980) is an ATPase subunit of the 26S proteasome regulatory complex, involved in ATP-dependent degradation of ubiquitinated proteins and in maintaining cellular homeostasis. The kit is provided in a 96-well microplate format and should be stored at 4°C for up to six months.
