The Assay Genie Progesterone Receptor Transcription Factor Activity Assay uses a microplate pre-coated with a consensus progesterone response element (PRE) oligonucleotide duplex. Activated progesterone receptor contained in nuclear lysates binds the immobilized DNA probe, and the captured complex is detected with a receptor-specific primary antibody followed by HRP-conjugated secondary reagent and a chromogenic substrate. The protocol completes in under four hours from loaded lysate to plate read, with a recommended sample volume of approximately 10 uL per well of freshly prepared nuclear extract. Reagent vials are packed with cold packs and should be stored at -20 degrees C; antibody components benefit from being aliquoted to limit freeze-thaw cycles, while the standard dilution buffer and wash concentrate remain stable at room temperature in the dark. The kit has been validated for reactivity against human, mouse, and rat progesterone receptor, with no significant cross-reactivity reported against closely related steroid receptors such as glucocorticoid receptor (GR) or estrogen receptor alpha (ER alpha), supporting selective measurement of PR-driven transcriptional activation in studies of hormone-responsive breast cancer models, endometrial signaling, and reproductive biology research use only.
