Description

From Assay Genie, this Cell-Based ELISA Kit is designed for the qualitative measurement of Progesterone Receptor phosphorylated at Ser-190 directly in cultured cells. It uses an indirect ELISA format in which target-specific primary antibodies capture the phospho-protein and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition.

The assay is reactive with human samples and is intended for use with adherent cells. Detection is read at 450 nm and it is reported with a dynamic range above 5000 cells per well. To support data normalization, a monoclonal anti-GAPDH antibody is included as an internal positive control, and a Crystal Violet whole-cell staining step can be used to account for differences in cell density across wells.

The target is encoded by the PGR gene (Gene ID: 5241; UniProt P06401), a member of the steroid hormone receptor superfamily. According to the UniProt annotation referenced in the product literature, phosphorylation at Ser-190 is among the basal phosphorylation sites on the receptor that is increased in response to progesterone and can be phosphorylated in vitro by the CDK2-A1 complex.
