Description

From Assay Genie, this cell-based ELISA kit is designed for detecting PRKAG1/2/3 protein expression in cultured adherent cells using a colorimetric method read at 450 nm. The assay uses an indirect ELISA format in which target-specific primary antibodies capture PRKAG1/2/3 and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition, allowing qualitative measurement of relative PRKAG1/2/3 levels. It can be used to measure the relative amounts of PRKAG1/2/3 in cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on PRKAG1/2/3.

A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and Crystal Violet whole-cell staining can be used to determine cell density and adjust for plating differences. Reactivity is reported for human, mouse, and rat samples, with a dynamic range of > 5000 cells. The kit is supplied in a 96-well microplate format and stored at 4°C for 6 months.

PRKAG1/2/3 are regulatory gamma subunits of the AMP-activated protein kinase (AMPK) complex, a conserved energy-sensing enzyme that monitors cellular energy status. Measurement of PRKAG1/2/3 activation supports research into cellular energy homeostasis, including regulation of glucose uptake, fatty acid oxidation, and mitochondrial biogenesis, and into the AMPK pathway's role in metabolic disorders, cancer, and neurodegenerative conditions.
