PPIF Colorimetric Cell-Based ELISA

Assay Genie offers a Cell-Based ELISA kit designed for the accurate detection of PPIF (Peptidylprolyl Isomerase F) levels in cell lysates and cell culture supernatants. The colorimetric assay enables quantification of PPIF across various cell types, with reactivity against human, mouse, and rat samples. PPIF, also known as cyclophilin D, plays a critical role in mitochondrial function and cell death pathways, and its dysregulation has been implicated in neurodegenerative disorders, cardiovascular diseases, and cancer.

The kit uses an indirect ELISA format in which PPIF is captured by PPIF-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. The bound HRP catalyses a colorimetric reaction upon substrate addition, read at 450 nm. The dynamic range is > 5000 Cells. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and Crystal Violet whole-cell staining is provided for determining cell density and adjusting for plating differences. The format is a 96-well microplate, and storage stability is 4°C for 6 months.
