The Assay Genie PPAR-gamma Transcription Factor Activity Assay is a non-radioactive DNA-binding immunoassay that quantifies activated PPAR-gamma binding to a plate-immobilized PPRE (PPAR response element) consensus oligonucleotide. The assay uses nuclear or whole-cell lysates, generally requiring 10 µL of nuclear extract per well, and reports relative transcription factor activity that reflects ligand activation by rosiglitazone, pioglitazone, 15-deoxy-delta-12,14-prostaglandin J2, and other receptor agonists. It is widely applied in adipogenesis, insulin signaling, metabolic disease, oncology, atherosclerosis, and chronic inflammation research. The kit ships with a pre-coated strip plate, binding buffer, positive control nuclear extract, HRP-conjugated detection antibody, wash buffer, TMB substrate, and stop solution. Validated reactivity covers human, mouse, and rat samples, supporting translational work across cell lines, primary cells, and tissue biopsies. Standard run time is approximately 3 hours, and the protocol yields a quantitative readout of DNA-binding activity that supports comparison of treatment conditions, dose-response stimulations, and time-course experiments.
