Description

From Assay Genie, this Cell-Based ELISA Kit is designed for the quantitative measurement of PPAR-gamma phosphorylated at serine 112 in cell lysates. The colorimetric assay is read at 450 nm and is validated for human, mouse, and rat reactivity, with a dynamic range of > 5000 cells. It is suited to adherent cell samples and offers a lysate-free, high-throughput workflow.

The kit uses an indirect ELISA format, in which PPAR-gamma (Phospho-Ser112) is captured by phospho-specific primary antibodies and detected via HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. It can be used to assess the relative levels of phosphorylated PPAR-gamma in cultured cells and to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on PPAR-gamma phosphorylation. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalization, and Crystal Violet whole-cell staining can be used to adjust for cell density. PPAR Gamma is a nuclear receptor involved in lipid metabolism, insulin sensitivity, and inflammation, and phosphorylation at Ser112 is a relevant target for research into metabolic disorders and diabetes.

Additional database identifiers associated with the target include Gene ID 5468 and UniProt ID P37231, with a reported molecular weight of 59.2 kD.
