PPAR-alpha Transcription Factor Activity Assay

This DNA-binding immunoassay from Assay Genie quantitatively measures the DNA-binding activity of activated PPAR-alpha in nuclear and whole-cell lysates. The microplate is pre-coated with immobilized oligonucleotides containing consensus Peroxisome Proliferator Response Elements (PPREs), enabling active PPAR-alpha dimers from human, mouse, or rat samples to bind and be detected with a PPAR-alpha-specific antibody in a colorimetric readout completed in roughly 3.5 hours.

The kit supports research into lipid metabolism, beta-oxidation of fatty acids, hepatic steatosis, NAFLD, and nuclear receptor signaling, and is frequently used to screen fibrate-class compounds and other candidate PPAR-alpha agonists or antagonists. Each kit provides a 96-well microplate, assay buffer, nuclear extraction reagents, detection antibody, and positive-control nuclear extract. Store kit components at -20 degrees C and protect from light to maintain antibody and oligonucleotide stability through the stated shelf life; thaw nuclear extracts on ice and limit freeze-thaw cycles. Load 20 uL of nuclear lysate per well for the standard protocol, with cross-reactivity validated for human, mouse, and rat PPAR-alpha.
