PP2A-alpha (Phospho-Tyr307)Colorimetric Cell-Based ELISA Kit

From Assay Genie, this cell-based ELISA kit enables qualitative measurement of PP2A-alpha phosphorylation at the Tyr307 site directly in cultured cells, without the need for lysate preparation. The assay uses an indirect ELISA format in which target-specific primary antibodies capture PP2A-alpha (Phospho-Tyr307), and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to drive a colorimetric reaction read at 450 nm.

Reactivity has been validated against human, mouse and rat samples, with a dynamic range of > 5000 Cells. A monoclonal GAPDH antibody is included as an internal positive control to normalize target absorbance, and a Crystal Violet whole-cell staining step can be used to determine cell density for further normalization. The kit is suitable for adherent cells and for screening the effects of treatments, siRNA, chemical inhibitors or activators on PP2A-alpha phosphorylation.

The target is encoded by the PPP2CA gene, a ubiquitous cytosolic Ser/Thr protein phosphatase catalytic subunit implicated in the regulation of cell growth, proliferation, differentiation and apoptosis, with phosphorylation at Tyr307 known to modulate its activity.
