PP2A-alpha Colorimetric Cell-Based ELISA Kit

From Assay Genie, this Cell-Based ELISA Kit is designed for the detection of PP2A-alpha protein expression profiles in cultured cells. The kit uses a colorimetric method with absorbance measured at 450 nm and is suited to adherent cells, providing a lysate-free, high-throughput workflow that can be used to measure relative target levels and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators.

Qualitative determination of PP2A-alpha is achieved through an indirect ELISA format, in which PP2A-alpha-specific primary antibodies capture the target while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody; the conjugated HRP then catalyses a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values. Following the colorimetric measurement, a Crystal Violet whole-cell staining method may be used to determine cell density, so that absorbance values can be normalised to cell amounts and plating differences adjusted.
