Description

A colorimetric cell-based ELISA kit from Assay Genie is designed for the accurate detection of potassium channel Kv3.2b levels in cell lysates and tissue homogenates. The kit is reactive with human, mouse, and rat samples and is intended for use with adherent cells in signal transduction research.

Detection is performed at 450 nm using an indirect ELISA format in which Kv3.2b-specific primary antibodies are bound by HRP-conjugated secondary antibodies, producing a colorimetric signal upon substrate addition. The assay has a dynamic range of > 5000 cells, is stored at 4 °C with a stated stability of 6 months, and is run in a microplate format. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density after the colorimetric measurement.

Potassium channel Kv3.2b, encoded by the KCNC2 gene (Gene ID 3747, UniProt Q96PR1), is a voltage-gated potassium channel that mediates the voltage-dependent potassium ion permeability of excitable membranes and plays a role in regulating neuronal excitability and action potential firing. The target is highlighted as a key area of interest for studying neurological disorders such as epilepsy, Parkinson's disease, and schizophrenia.
