Description

From Assay Genie, this sandwich double-antibody ELISA kit enables quantitative measurement of porcine VEGFR1/Flt1 (Vascular Endothelial Growth Factor Receptor 1) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The microplate is pre-coated with a capture antibody specific to the target. Standards and appropriately diluted samples are added to the wells and incubated, allowing the analyte to bind, followed by a biotinylated detection antibody that forms a sandwich complex. HRP-conjugated Streptavidin is added, a colorimetric reaction is developed with TMB substrate, and the reaction is stopped with an acidic solution before absorbance is measured at 450 nm.

Sensitivity is 18.75 pg/mL with a quantitative range of 31.25–2000 pg/mL. The kit should be stored at 2-8°C for 12 months. Linearity (n = 5) for serum at 1:2, 1:4 and 1:8 dilutions is 85-105%, 84-99% and 80-100%; for EDTA plasma 85-105%, 86-101% and 80-99%; and for heparin plasma 86-101%, 83-100% and 81-95%. Recovery averages are 96% for serum, 90% for EDTA plasma and 97% for heparin plasma. The associated UniProt identifier is F1RSU5, and reactivity is reported as porcine (swine).

The protocol includes equilibrating reagents and TMB substrate to room temperature, primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody binding for 60 minutes, HRP-Streptavidin (SABC) binding for 30 minutes, TMB color development in the dark for 10–20 minutes, and a stop step before reading at 450 nm.
