Transcription factor AP-1 (JUN), also known as c-Jun, is a central regulator of gene expression involved in cell proliferation, differentiation, apoptosis, and stress response pathways. This Assay Genie sandwich ELISA kit provides a quantitative, colorimetric detection method for measuring JUN protein levels in porcine serum and plasma samples.

The assay utilizes pre-coated microplate wells with capture antibodies specific to porcine JUN, paired with biotinylated detection antibodies and streptavidin-HRP conjugate for signal generation. Results are read on a standard microplate reader at 450 nm wavelength. Each kit includes all necessary reagents for 96 determinations, with a standard curve for accurate quantification.

Store reagents at 2-8 degrees C; allow all components to reach room temperature before use. For research use only, not intended for diagnostic procedures.

AP-1 complexes formed by c-Jun together with c-Fos bind to the TPA response element (TRE) and regulate transcription of genes involved in the cell cycle, apoptosis, and inflammation. Phosphorylation of c-Jun by JNK on serine residues (including Ser63 and Ser73) enhances its transcriptional activity, and these signaling events are routinely examined in oxidative stress, liver injury, and oncogenesis studies.

This porcine-reactive Assay Genie kit is commonly used in cardiovascular, hepatic, and reproductive biology research, where the pig serves as a translational model. Each microplate well typically accommodates 100 µL of sample, with an assay range and standard curve calibrated against recombinant porcine JUN. Reagents are stable for up to 6 months at 2-8 °C when stored unopened, and the full workflow generally completes in under 4 hours from sample loading to absorbance reading at 450 nm.

For best results, avoid repeated freeze-thaw cycles of serum or plasma aliquots and equilibrate all components to room temperature (20-25 °C) before pipetting. Researchers often pair this ELISA with western blot or immunohistochemistry to confirm JUN expression changes in porcine liver, myocardium, or endometrial tissues.
