Description

From Assay Genie, this Porcine TIMP3 (Metalloproteinase inhibitor 3) ELISA Kit is a sandwich-format, double-antibody assay intended to quantify TIMP3 in porcine samples. The kit is suited to serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and uses colorimetric detection. TIMP3 is described as a key regulator of metalloproteinases, with dysregulation implicated in cancer, cardiovascular disorders and inflammatory conditions.

Reported sensitivity is 0.375 ng/mL with an assay range of 0.625-40 ng/mL. Spiked recovery averaged 92% (range 87-98%) for serum, 95% (range 87-102%) for EDTA plasma and 96% (range 86-104%) for heparin plasma (n=5 each). Linearity on 1:2, 1:4 and 1:8 dilutions was 89-98%, 85-103% and 88-105% for serum; 83-94%, 83-99% and 83-101% for EDTA plasma; and 80-96%, 85-100% and 82-94% for heparin plasma (n=5 each).

The workflow involves equilibrating reagents and loading standards and samples for incubation at 37°C for 90 minutes, addition of a biotin-labelled detection antibody (37°C for 60 minutes), HRP-Streptavidin (37°C for 30 minutes), TMB substrate in the dark for 10-20 minutes, and reading absorbance at 450 nm after stop solution. Serum is processed by clotting and centrifugation at 1000 × g for 20 minutes; plasma is collected with anticoagulant and centrifuged at 1000 × g for 15 minutes at 2-8°C. The kit should be stored at 2-8°C for up to 12 months.
