Porcine SLIT2/Slit homolog 2 ELISA Kit

This Sandwich ELISA from Assay Genie is designed for the precise measurement of SLIT2 (Slit Homolog 2) levels in porcine samples. It employs a double-antibody sandwich format with colorimetric detection and is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit supports research into SLIT2, a protein involved in neuronal development and axon guidance that contributes to the regulation of cell migration and tissue morphogenesis, and that has been implicated in various neurological disorders.

According to the manufacturer's specifications, the assay has a sensitivity of 0.188 ng/mL and a detection range of 0.313-20 ng/mL. Reported linearity (n=5) at 1:2, 1:4 and 1:8 dilutions is 89-105%, 93-104% and 92-99% for serum; 89-98%, 82-100% and 88-100% for EDTA plasma; and 80-100%, 81-97% and 81-93% for heparin plasma. Recovery (n=5) is 86-101% (average 95%) for serum, 89-104% (average 98%) for EDTA plasma, and 88-104% (average 99%) for heparin plasma. All kit components should be stored at 2-8°C, with a stated shelf life of 12 months under those conditions.

The procedure includes: equilibrating reagents and TMB substrate to room temperature, loading standards, samples and blanks on the pre-coated plate and incubating at 37°C for 90 minutes; adding biotin-labelled detection antibody and incubating at 37°C for 60 minutes; adding HRP-Streptavidin (SABC) and incubating at 37°C for 30 minutes; developing with TMB substrate in the dark for 10-20 minutes; and adding stop solution before reading absorbance at 450 nm. Supplied components comprise a dismountable ELISA microplate, lyophilized standard, biotin-labelled antibody (100X), HRP-Streptavidin conjugate (SABC, 100X), TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, stop solution, 25X wash buffer, and plate sealer. The protocol is batch- and lot-specific, so the manual supplied with the kit should be followed.
