Description
This Sandwich ELISA kit from Assay Genie is designed for the precise and reliable detection of Placenta Growth Factor (PIGF) in porcine samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. It uses a colorimetric detection method. PIGF is a key protein involved in angiogenesis, promoting the growth of blood vessels and influencing cell proliferation, and its involvement in cancer, cardiovascular diseases and neurodegenerative disorders makes it a valuable biomarker for biomedical research.
The assay uses a Sandwich ELISA, Double Antibody format with a sensitivity of 18.75 pg/mL and an assay range of 31.25-2000 pg/mL. Reagents are stored at 2-8°C for 12 months. The procedure includes equilibrating reagents and TMB substrate to room temperature, a primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin (SABC) binding at 37°C for 30 minutes, colour development with TMB substrate in the dark for 10-20 minutes, and a stop reaction with absorbance read at 450 nm.
Linearity across 1:2, 1:4 and 1:8 dilutions was 91-102%, 88-100% and 84-96% for serum; 87-105%, 85-99% and 82-100% for EDTA plasma; and 87-105%, 82-96% and 81-96% for heparin plasma (n=5 each). Recovery was 87-101% (mean 94%) for serum, 88-101% (mean 93%) for EDTA plasma, and 87-98% (mean 92%) for heparin plasma (n=5 each).
