Porcine PAI1/Plasminogen activator inhibitor 1 ELISA Kit

Designed for the quantitative measurement of Plasminogen Activitor Inhibitor-1 (PAI-1) in porcine samples, this Sandwich ELISA kit from Assay Genie delivers high sensitivity and specificity for research applications. PAI-1 is a critical regulator of the fibrinolytic pathway, inhibiting tissue plasminogen activator and urokinase and influencing thrombosis and hemostasis. Dysregulation of PAI-1 has been linked to cardiovascular diseases, diabetes, and fibrotic disorders, making it a valuable biomarker for study.

The assay uses colorimetric detection with a double-antibody sandwich format and is compatible with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Performance specifications include a sensitivity of 0.188 ng/mL and a measurable range of 0.313-20 ng/mL. Unopened kits are stored at 2-8°C for 12 months. Linearity across 1:2, 1:4, and 1:8 dilutions (n=5 each) was 84-100%, 82-100%, and 84-98% for serum; 85-100%, 87-104%, and 86-103% for EDTA plasma; and 91-102%, 83-98%, and 82-95% for heparin plasma. Recovery averaged 94% in serum (range 91-103%), 92% in EDTA plasma (range 92-100%), and 96% in heparin plasma (range 89-105%).

The workflow involves equilibrating reagents and loading standards, samples, and controls onto the pre-coated plate, incubating at 37°C for 90 minutes, adding biotin-labelled detection antibody for 60 minutes, binding HRP-Streptavidin for 30 minutes, developing with TMB substrate in the dark for 10-20 minutes, and reading absorbance at 450 nm after stop solution addition.
