Orexin (also called hypocretin, HCRT) is a neuropeptide synthesized by neurons in the lateral hypothalamus that regulates wakefulness, feeding behavior, energy homeostasis, and reward pathways. Dysregulation of orexin signaling has been implicated in porcine models of narcolepsy, sleep fragmentation, appetite control, and metabolic disorders, making quantitative measurement valuable for both agricultural and biomedical research.

This Sandwich ELISA from Assay Genie uses a colorimetric detection method on a 96-well microplate and is validated for porcine serum and plasma samples. Standard curve concentrations typically span roughly 0.1 to 10 ng/mL, with sensitivity sufficient for detecting endogenous orexin levels in circulation. The assay follows a standard sandwich immunoassay workflow: capture antibody pre-coated wells, add sample and biotinylated detection antibody, wash, incubate with streptavidin-HRP, then develop with TMB substrate and read absorbance at 450 nm.

Store all kit components at 2-8 °C unless otherwise indicated on the vial, and bring reagents to room temperature before use. Allow samples and standards to equilibrate, and avoid repeated freeze-thaw cycles to preserve peptide integrity. For reproducible results, run samples in duplicate and generate a fresh standard curve on each plate. Intended for research use only.
