Description

A colorimetric sandwich ELISA from Assay Genie designed for the quantitative detection of Osteoprotegerin (OPG) in porcine samples, including serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples. Osteoprotegerin is described as a key regulator of bone metabolism and bone homeostasis, and altered OPG levels have been associated with bone-related disorders such as osteoporosis and osteoarthritis, making the kit a useful tool for bone biology research and studies of skeletal disorders in porcine models.

The kit uses a double-antibody sandwich format with a sensitivity of 18.75 pg/mL and a working range of 31.25-2000 pg/mL. Absorbance is read at 450 nm following TMB substrate development. Linearity has been demonstrated in serum (92-97% at 1:2, 84-101% at 1:4, 81-99% at 1:8), EDTA plasma (85-103%, 87-95%, 88-96%) and heparin plasma (93-104%, 83-101%, 82-100%), while spike-and-recovery averages are 97% in serum, 95% in EDTA plasma, and 93% in heparin plasma. The standard assay procedure involves primary incubation at 37°C for 90 minutes, detection antibody binding for 60 minutes, and HRP-streptavidin binding for 30 minutes, with TMB colour development for 10-20 minutes in the dark. Unopened kits should be stored at 2-8°C and remain stable for 12 months under these conditions.
