Description

Assay Genie's Porcine Neuropeptide Y ELISA is a sandwich-format, double-antibody ELISA kit designed to quantify neuropeptide Y (NPY) in porcine samples. Reactivity is specific to porcine species, and the assay uses a colorimetric detection method. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit reports a sensitivity of 18.75 pg/mL and a working range of 31.25–2000 pg/mL. Reagents are stored at 2–8°C for up to 12 months. According to UniProt entry P01304, NPY is implicated in the control of feeding and in the secretion of gonadotrophin-release hormone.

Reported linearity at 1:2, 1:4 and 1:8 dilutions (n = 5) is 82–100%, 81–100%, and 85–97% for serum; 82–98%, 82–92%, and 85–105% for EDTA plasma; and 88–100%, 87–101%, and 82–99% for heparin plasma. Recovery ranges (with averages) are 87–103% (90%) for serum, 85–100% (93%) for EDTA plasma, and 85–105% (90%) for heparin plasma. The protocol calls for a 90-minute primary incubation at 37 °C, a 60-minute biotin-labelled detection antibody step, a 30-minute HRP-Streptavidin binding step, TMB colour development in the dark for 10–20 minutes, and immediate absorbance measurement at 450 nm after stop solution is added. Sample preparation guidance covers serum clotting, plasma centrifugation with anticoagulant, tissue homogenisation in PBS with protease inhibitors, cell culture supernatant clarification, and cell lysate preparation.
