Description

A colorimetric sandwich ELISA from Assay Genie designed for the quantitative detection of porcine Netrin-1. The kit uses a double-antibody sandwich format with a reported sensitivity of 37.5 pg/mL and a measurable range of 62.5-4000 pg/mL. It is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and unopened kits are stored at 2-8°C for up to 12 months.

Reported linearity in serum (n=5) spans 91-105% at 1:2, 88-104% at 1:4 and 88-101% at 1:8; EDTA plasma (n=5) spans 84-100%, 84-97% and 86-101% across the same dilutions; heparin plasma (n=5) spans 81-98%, 82-97% and 80-99%. Spike-and-recovery averages are 92% for serum, 92% for EDTA plasma and 94% for heparin plasma, with corresponding recovery ranges of 88-98%, 85-101% and 85-105%. The workflow is a six-step sandwich ELISA: equilibrate reagents and the TMB substrate, load standards and samples for a 90-minute antigen-binding incubation at 37°C, add biotin-labelled detection antibody for 60 minutes at 37°C, add HRP-streptavidin (SABC) for 30 minutes at 37°C, develop colour with TMB in the dark for 10-20 minutes, then add stop solution and read absorbance at 450 nm.

Netrin-1 is a protein implicated in axon guidance and neuronal development, and its dysregulation has been linked to neurological disorders, making this kit suited to research into neurodevelopmental processes and related disease mechanisms.
