Description

A sandwich ELISA from Assay Genie intended for the quantitative detection of porcine MIP-1 beta (Macrophage Inflammatory Protein-1 beta), a chemokine that plays a role in the inflammatory response by recruiting immune cells to sites of infection or tissue damage. The kit uses a double-antibody sandwich format with colorimetric readout and is suitable for use with porcine serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

Reported performance characteristics include a sensitivity of 18.75 pg/mL and a working range of 31.25–2000 pg/mL. Recovery across serum, EDTA plasma and heparin plasma averaged 98%, 92% and 93% respectively, with dilution linearity in the same matrices falling between 80% and 105%. The kit is supplied with a pre-coated dismountable microplate, lyophilized standard, biotin-labelled detection antibody, HRP-streptavidin conjugate, TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, stop solution, wash buffer and plate sealer, and components should be stored at 2–8°C for up to 12 months.

The assay procedure consists of equilibrating reagents and loading standards, samples and controls onto the pre-coated plate for a 90-minute primary incubation at 37°C, a 60-minute biotin-labelled detection antibody incubation, a 30-minute HRP-streptavidin (SABC) binding step, TMB colour development in the dark for 10–20 minutes, and a stop reaction with absorbance read at 450 nm. Elevated MIP-1 beta levels have been implicated in various inflammatory conditions, making the kit a useful tool for studying immune responses in porcine models.
