Description

The Assay Genie Porcine Luteinizing Hormone ELISA Kit is a competitive ELISA coated with antigen, intended for quantitative measurement of luteinizing hormone in porcine samples. Accepted sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit carries the UniProt reference P01232 and is suitable for research into porcine reproductive physiology, fertility, and breeding programme management.

Reported performance figures include a sensitivity of 0.469 ng/mL and a detection range of 0.781–50 ng/mL. Linearity in serum (n=10) was 88–97% at 1:2, 82–99% at 1:4, and 84–95% at 1:8 dilution; EDTA plasma (n=10) showed 91–104%, 84–97% and 80–100% at the same dilutions; heparin plasma (n=10) gave 86–101%, 82–100% and 80–98%. Recovery averaged 91% for serum, 95% for EDTA plasma and 98% for heparin plasma across the respective ranges of 85–103%, 90–104% and 91–104%. Reagents are stored sealed at 2–8°C and must not be cryopreserved.

The protocol involves adding 50 µL of standard or sample followed by 50 µL of biotin-labelled antibody per well, incubating at 37°C for 45 minutes, washing the plate five times, then developing with TMB substrate in the dark at 37°C for 10–20 minutes before stopping the reaction and reading absorbance at 450 nm. Sample preparation guidance covers serum, plasma, cell culture supernatant, cell lysate, tissue homogenate and other liquid samples. The kit has been cited in a study by Batorek-Lukač et al. in Andrology 2022 (PubMed ID: 35752946) on immunocastration in adult boars as a model for late-onset hypogonadism.
