The Porcine Inhibin alpha chain (INHA) ELISA Kit from Assay Genie uses a quantitative sandwich immunoassay format on a pre-coated 96-well microplate, with colorimetric detection measured at 450 nm and validated for serum and plasma samples. The kit supports reproductive biology research, including studies of folliculogenesis, gonadal steroidogenesis, and FSH regulatory feedback in swine models. Standard assay time is approximately 3 hours at room temperature, and results are read on a standard microplate reader. Store reagents at 2-8°C, protect from light, and avoid repeated freeze-thaw cycles of the working conjugate to preserve performance through the stated shelf life. Intended for research use only.

Target Biology: Inhibin Alpha Chain (INHA)

Inhibin alpha chain is the common subunit of inhibin A and inhibin B, heterodimeric glycoproteins secreted by ovarian granulosa cells and testicular Sertoli cells. In swine, circulating inhibin suppresses pituitary FSH release, and serum alpha-subunit levels serve as a biomarker of follicular reserve, estrus cyclicity, and gonadal pathology. The Assay Genie sandwich ELISA detects both free alpha chain and dimeric inhibin forms, supporting work on granulosa cell tumors, cryptorchidism, and litter-size selection in sows.

Assay Workflow

The pre-coated 96-well microplate is supplied as 12 removable 8-well strips. Standards, controls, and serum or plasma samples are pipetted into wells, followed by a biotinylated detection antibody and streptavidin-HRP conjugate. After TMB substrate development, stop solution is added and absorbance is read at 450 nm, with 630 nm optional reference correction. Typical standard curve range is 15.6 to 1000 pg/mL, with intra-assay CV below 10% and inter-assay CV below 15%.

Kit Contents & Storage

The kit provides a pre-coated microplate, lyophilized recombinant porcine INHA standards, biotinylated detection antibody, streptavidin-HRP conjugate, 30 mL wash buffer concentrate, TMB substrate, stop solution, and adhesive plate seals. Unopened reagents are stable for up to 6 months at 2-8 °C; reconstituted standards and diluted working conjugate should be aliquoted and stored at -20 °C to limit freeze-thaw damage. Equilibrate all components to room temperature before use and keep TMB protected from light. For research use only.
