Porcine IL-1RA/IL-1 receptor alpha ELISA Kit

The Assay Genie kit for porcine IL-1RA / IL-1 receptor alpha is a sandwich ELISA in a double-antibody format with a colorimetric detection method. It is intended for the quantitative measurement of IL-1RA / IL-1 receptor alpha in porcine samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

The assay has a sensitivity of 37.5 pg/mL and a standard range of 62.5-4000 pg/mL. Components should be stored at 2-8°C for 12 months. Validated linearity for serum (n = 5) is 98-102% at 1:2, 90-105% at 1:4, and 85-103% at 1:8; for EDTA plasma (n = 5) 84-94% at 1:2, 86-91% at 1:4, and 89-101% at 1:8; and for heparin plasma (n = 5) 80-100% at 1:2, 81-96% at 1:4, and 82-100% at 1:8. Recovery in serum (n = 5) is 87-103% with an average of 92%; in EDTA plasma (n = 5) 88-100% with an average of 94%; and in heparin plasma (n = 5) 89-105% with an average of 97%.

The procedure involves equilibrating reagents to room temperature, loading standards, samples and blanks, then incubating the plate at 37°C for 90 minutes. A biotin-labelled detection antibody is added and incubated for 60 minutes at 37°C, followed by HRP-Streptavidin (SABC) for 30 minutes. TMB substrate is then added and incubated in the dark for 10-20 minutes, after which stop solution is added and absorbance is read at 450 nm.
