Description

A sandwich ELISA from Assay Genie enables the quantitative measurement of insulin-like growth factor 1 (IGF-1) in porcine samples. The double antibody format is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The reported sensitivity is 0.188 ng/mL with a working range of 0.313-20 ng/mL. Reagents are stored at 2-8°C and the kit is stated to remain stable for 12 months under these conditions.

According to the product literature, IGF-1 is a key regulator of growth and development in animals, with roles in cell proliferation, differentiation, survival, and metabolic regulation, and serves as a biomarker for research into metabolic diseases, growth disorders, and muscle development. The protocol comprises a primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding for 60 minutes, HRP-streptavidin binding for 30 minutes, TMB substrate colour development in the dark for 10–20 minutes, and absorbance measurement at 450 nm after addition of stop solution. Sample preparation guidelines cover serum, plasma, tissue homogenate, cell culture supernatant, and cell lysate.

Linearity has been evaluated at 1:2, 1:4, and 1:8 dilutions (n=5 per matrix): serum 86-98%, 82-99%, and 86-100%; EDTA plasma 86-103%, 83-94%, and 84-97%; heparin plasma 85-101%, 82-96%, and 80-93%. Average recovery was 96% for serum, 90% for EDTA plasma, and 96% for heparin plasma (n=5). The intended research areas include animal physiology, growth and development, and metabolic disorders.
