Description

From Assay Genie, this Sandwich ELISA kit is designed for the quantitative measurement of IgE in porcine serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. It uses a double-antibody sandwich format with colorimetric detection and is intended for research applications investigating porcine allergic responses and immune function.

Reported performance characteristics include a sensitivity of 0.188 ng/mL and a standard range of 0.313–20 ng/mL. Linearity has been evaluated at 1:2, 1:4 and 1:8 dilutions in serum, EDTA plasma and heparin plasma (each n = 5), and spike-and-recovery has been assessed in the same three matrices, supporting reliable performance across common porcine sample types. The workflow uses a pre-coated plate on which standards, samples and controls are loaded for a 37 °C primary incubation of 90 minutes, followed by incubation with a biotin-labelled detection antibody for 60 minutes and HRP-Streptavidin (SABC) for 30 minutes, both at 37 °C. TMB substrate is then added for 10–20 minutes in the dark, the reaction is stopped, and absorbance is read at 450 nm.

Sample preparation guidance is provided for serum (allow blood to clot, then centrifuge at 1000 × g for 20 minutes), plasma (collect using anticoagulant tubes, centrifuge at 1000 × g for 15 minutes at 2–8 °C), tissue homogenate, cell culture supernatant (centrifuge at 2500 rpm for 5 minutes) and cell lysate workflows. Unopened kit components are stored at 2–8 °C for 12 months.
