Built around a quantitative sandwich ELISA workflow, this Porcine Hemoglobin subunit alpha (HBA) ELISA Kit from Assay Genie enables sensitive measurement of HBA in porcine serum and plasma. Hemoglobin subunit alpha pairs with subunit beta to form the alpha2beta2 tetramer that carries oxygen in red blood cells, so tracking circulating HBA supports swine models of anemia, hemorrhage, blood loss, transfusion compatibility, erythropoiesis, and cardiovascular function. The assay is run on a pre-coated 96-well microplate and produces a colorimetric signal read at 450 nm on a standard plate reader. The kit ships with capture antibody, biotinylated detection antibody, recombinant standards, streptavidin-HRP conjugate, wash buffer, substrate, and stop solution. Store unopened kits refrigerated at 2-8 degrees C, keep standards and conjugate frozen at -20 degrees C for longer shelf life, allow reagents to equilibrate to room temperature before use, and limit freeze-thaw cycles of the standard to preserve performance. For research use only.

Operating as a quantitative sandwich immunoassay from Assay Genie, the assay uses a pre-coated 96-well microplate (8 x 12 dismountable strips), a biotinylated detection antibody, and streptavidin-HRP to convert HBA binding into a measurable colorimetric signal at 450 nm. The standard protocol adds 100 µL of sample, standard, or blank per well, incubates for 120 minutes at 37 degrees C, then sequentially introduces Detection Reagent A (1 hour at 37 degrees C), washes three times with approximately 400 µL of wash buffer, introduces Detection Reagent B (60 minutes at 37 degrees C), washes five times, develops with 90 µL of substrate for 10-20 minutes at 37 degrees C protected from light, and stops the reaction with 50 µL of stop solution before reading optical density. Serum samples should clot 30 minutes at room temperature and be centrifuged at 1,000 x g for 10 minutes, while plasma collected on EDTA or heparin is centrifuged at 1,000 x g for 15 minutes at 4 degrees C within 30 minutes of draw; aliquots are best held at -80 degrees C with freeze-thaw cycles minimized. The supplied kit contains the pre-coated microplate, two vials of lyophilized recombinant standard, sample diluent (20 mL), assay diluents A and B (10 mL each), detection reagents A and B (120 µL each), wash buffer (30 mL), TMB substrate (10 mL), stop solution (10 mL), and plate sealers. Unopened kit components are stored refrigerated at 2-8 degrees C with standards and detection reagents held at -20 degrees C, and all reagents should be equilibrated to room temperature before use. Supports research applications in swine models of erythropoiesis, anemia, hemorrhage, transfusion compatibility, and cardiovascular pathophysiology, with intra-assay and inter-assay CV, linearity, and recovery metrics provided in the lot-specific certificate of analysis. For research use only.
