Quantitative measurement of ghrelin (GHRL) in porcine samples is achieved through this competitive immunoassay from Assay Genie, applied in research on appetite regulation, energy homeostasis, growth hormone release, and gastrointestinal motility in swine models.

The plate handles 96 assays in a single run, accommodating diverse research matrices including tissue homogenates, cell lysates, cell culture supernatants, and circulatory specimens. Absorbance-based readout requires only a standard microplate reader for quantification.

Ghrelin is a 28-amino acid orexigenic peptide produced chiefly by P/D1-type cells in the oxyntic mucosa of the stomach. It acts as the endogenous ligand for the growth hormone secretagogue receptor 1a (GHS-R1a), stimulating pituitary GH release and influencing insulin secretion, glucose homeostasis, gastric acid secretion, and short-term energy balance. The competitive format accommodates both acyl-ghrelin, the active Ser3-octanoylated form, and total circulating ghrelin when paired with appropriate sample handling.

Recommended sample preparation includes protease inhibitor cocktails for tissue lysates and rapid cooling with optional acidification of circulatory specimens when active acyl-ghrelin is the analyte. A standard wash-and-read workflow applies once samples are prepared, with absorbance values inversely proportional to ghrelin concentration in the test specimen.
