Description

From Assay Genie, this Porcine EPO (Erythropoietin) ELISA Kit is a sandwich ELISA, double antibody assay designed for the precise measurement of erythropoietin in porcine samples. It is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with a reported sensitivity of 18.75 pg/mL and a measuring range of 31.25-2000 pg/mL. Detection is colorimetric. Erythropoietin is described in the product literature as a vital hormone that stimulates red blood cell production and is essential for oxygen delivery, making it a valuable biomarker for studying conditions such as anemia, kidney disease, and chronic illnesses.

Reagents are stored at 2-8°C for 12 months. The assay procedure involves equilibrating reagents and TMB substrate to room temperature, setting standard and test wells on the pre-coated plate, primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin binding at 37°C for 30 minutes, color development in the dark for 10–20 minutes, and absorbance reading at 450 nm after stop solution. Reported linearity across dilutions of 1:2, 1:4 and 1:8 is 85-104%, 82-101%, 81-95% for serum; 90-104%, 83-101%, 85-96% for EDTA plasma; and 87-103%, 89-101%, 82-100% for heparin plasma. Recovery ranges are 87-97% (average 92%) for serum, 86-97% (average 92%) for EDTA plasma, and 86-99% (average 93%) for heparin plasma.

Sample preparation guidance includes allowing blood to clot and centrifuging at 1000 × g for 20 minutes for serum, collecting plasma with anticoagulant tubes and centrifuging at 1000 × g for 15 minutes at 2-8°C, and centrifuging cell culture supernatant at 2500 rpm for 5 minutes. Synonyms listed for the target include EPO, Erythropoietin, EP, MVCD2, Epoetin, Hematopoietin, and Hemopoietin, with Uniprot reference P49157.
