Description
The Assay Genie Porcine Diacylglycerol kinase alpha (DGKA) ELISA Kit is a sandwich-format enzyme-linked immunosorbent assay designed for the quantitative measurement of DGKA in porcine samples. The kit uses a 96-well microtiter plate pre-coated with an antibody specific to DGKA. A biotinylated detection antibody specific for DGKA and an Avidin-Horseradish Peroxidase (HRP) conjugate are added sequentially, and the resulting enzymatic reaction is terminated with a sulphuric acid solution. Optical density is read spectrophotometrically at a wavelength of 450 nm ± 2 nm, with values proportional to DGKA concentration and quantifiable against a standard curve.
Compatible sample types include serum, plasma, saliva, urine, cell culture supernatant, and tissue homogenates, and the assay is suitable for routine laboratory and high-throughput screening workflows. Serum is collected via a serum separator tube, allowed to clot for 30 minutes, then centrifuged for 15 minutes at approximately 1000 × g. Plasma is collected using EDTA or heparin as anticoagulant and centrifuged for 15 minutes at 1000 × g at 2-8°C. Urine is centrifuged for 20 minutes at 2000-3000 rpm; a similar protocol applies to cerebrospinal fluid. Tissue samples are rinsed with 1× PBS, homogenized, subjected to two freeze-thaw cycles, and centrifuged for 5 minutes at 5000 × g. Samples containing NaN3 or showing haemolysis are not suitable. The kit contains an Assay Plate, Standard, Sample Diluent, Assay Diluent A, Assay Diluent B, Detection Reagent A, Detection Reagent B, Wash Buffer concentrate, Substrate, Stop Solution, and Plate Sealers. The Assay Plate, Standard, and Detection Reagents A and B are stored at -20°C and the Substrate at 4°C; for long-term storage the kit (excluding the Substrate) is kept at -20°C. The test kit is stable for six months from the date of manufacture when stored as prescribed, and samples may be kept at 2-8°C for up to 5 days or stored at -20°C or -80°C for longer periods to avoid loss of bioactivity.
