Porcine D-amino-acid oxidase/DAO ELISA Kit

From Assay Genie, this Sandwich ELISA kit is designed for the quantitative measurement of D-amino-acid oxidase (DAO) in porcine samples. The assay uses a double-antibody sandwich format with colorimetric readout, and is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

The kit reports a sensitivity of 0.938 ng/mL and an assay range of 1.563-100 ng/mL. Linearity was assessed at 1:2, 1:4 and 1:8 dilutions (n=5): serum gave 82-100%, 81-100% and 90-98%; EDTA plasma gave 82-98%, 82-92% and 85-105%; heparin plasma gave 85-100%, 87-101% and 82-99%. Spike-and-recovery testing returned average values of 95% for serum, 93% for EDTA plasma and 90% for heparin plasma.

The procedure begins with reagent and plate equilibration, followed by a primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin incubation at 37°C for 30 minutes, TMB colour development in the dark for 10-20 minutes, and a stop reaction with absorbance read at 450 nm. Unopened reagents should be stored at 2-8°C for 12 months.
