CXCL16 is a distinctive chemokine that exists in both a transmembrane form and a cleaved soluble form, signaling through the CXCR6 receptor to drive lymphocyte recruitment, scavenger receptor activity, and localized inflammation in swine. This Sandwich ELISA from Assay Genie quantifies porcine CXCL16 in serum and plasma on a pre-coated 96-well microplate, producing a colorimetric signal read at 450 nm with a microplate reader. Reagents are shipped refrigerated and should be stored at 2-8 °C; reconstituted standards and detection antibody vials remain stable through limited freeze-thaw cycles when aliquoted. The 96-well plate format supports duplicate measurements of up to 40 unknown samples per run alongside a standard curve, and the assay is validated for in vitro research use only in studies of infectious disease, vascular inflammation, hepatic fibrosis, and reproductive immunology in pigs.

The Assay Genie workflow runs in approximately 3.5 to 4 hours at room temperature once samples are equilibrated, with all incubations performed on the supplied adhesive plate sealers to prevent evaporation. The plate is supplied as 12 removable 8-well strips in a sealed foil pouch with desiccant, so unused strips can be returned and used within the kit shelf life. A typical standard curve spans 7 dilution points from roughly 15 pg/mL to 1000 pg/mL of recombinant porcine CXCL16, with intra-assay and inter-assay coefficients of variation generally below 10%. Recommended sample volume is 50 µL per well for serum and plasma, with samples diluted in the supplied assay diluent when concentrations fall outside the linear range of the curve. The kit specifically recognizes porcine CXCL16 and shows no significant cross-reactivity with related chemokines. Reagent vials should be returned to 2-8 °C immediately after use, and the pre-coated microplate, biotinylated detection antibody, streptavidin-HRP conjugate, TMB substrate, stop solution, and 20× wash buffer concentrate remain stable under these conditions through the printed expiry. Porcine models in which this marker is commonly profiled include PRRSV-challenged alveolar macrophages, CCl4- and alcohol-induced hepatic fibrosis, atherosclerotic lesion development in hyperlipidemic minipigs, and endometrial remodeling during early gestation in gilts and sows.
