Description

A sandwich ELISA from Assay Genie designed for the quantitative measurement of basic fibroblast growth factor (bFGF/FGF2) in porcine samples. The assay uses a colorimetric, double-antibody sandwich format and is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate and other liquid samples.

The kit offers a sensitivity of 9.375 pg/mL and a working range of 15.625-1000 pg/mL. Linearity for serum (n=5) spans 80-105% across 1:2, 1:4 and 1:8 dilutions, EDTA plasma (n=5) spans 82-104%, and heparin plasma (n=5) spans 82-99%. Mean recovery is 97% in serum, 99% in EDTA plasma and 92% in heparin plasma. Reagents are stored at 2-8°C for 12 months.

Basic fibroblast growth factor (bFGF/FGF2) is a key growth factor implicated in cell growth, proliferation and differentiation, and in processes including wound healing, tissue regeneration and embryonic development; it is also linked to the pathogenesis of cancer and cardiovascular disorders, making it a useful biomarker for disease research. The workflow involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples and a zero control onto the pre-coated plate, a 90-minute primary incubation at 37°C, addition of a biotin-labelled detection antibody for 60 minutes, HRP-Streptavidin binding for 30 minutes, TMB colour development in the dark for 10-20 minutes, and absorbance reading at 450 nm after adding the stop solution. Sample-handling guidance covers serum (clot, centrifuge at 1000 × g for 20 minutes), plasma (collect with anticoagulant, centrifuge at 1000 × g for 15 minutes at 2-8°C), tissue homogenates (PBS with protease inhibitors), cell culture supernatant (centrifuge at 2500 rpm for 5 minutes) and cell lysates (lysis buffer with protease inhibitors).
