Description

Assay Genie's Porcine ANP (Atrial Natriuretic Peptide) Converting Enzyme ELISA Kit is a sandwich, double-antibody immunoassay designed to measure ANP converting enzyme levels in porcine samples. The ANP converting enzyme plays a critical role in the regulation of blood pressure and fluid balance by converting ANP into biologically active forms, and the kit is positioned for cardiovascular physiology and disease-mechanism research in porcine models. Detection is colorimetric and the kit is supplied with a pre-coated ELISA microplate together with the reagents needed to run the protocol.

The assay is compatible with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported performance specifications include a sensitivity of 9.375 pg/mL and a range of 15.625-1000 pg/mL. Linearity across 1:2, 1:4, and 1:8 dilutions is reported as 91-100%, 88-103%, and 86-103% for serum (n = 5); 86-105%, 85-102%, and 84-101% for EDTA plasma (n = 5); and 87-100%, 81-96%, and 84-100% for heparin plasma (n = 5). Recovery ranges and averages are 89-103% (average 96%) for serum, 93-103% (average 97%) for EDTA plasma, and 88-105% (average 98%) for heparin plasma. Unopened kits are stored at 2-8°C for 12 months.

The procedure involves equilibrating reagents and TMB substrate to room temperature, loading standards, samples and blanks onto the pre-coated plate, and incubating at 37°C for 90 minutes for primary antigen binding. A biotin-labeled detection antibody is then added and incubated at 37°C for 60 minutes, followed by HRP-Streptavidin (SABC) at 37°C for 30 minutes. TMB substrate is added and incubated in the dark for 10-20 minutes before the reaction is stopped and absorbance is read at 450 nm. Sample-preparation guidance is provided for serum, plasma, tissue homogenate, cell culture supernatant, cell lysate, and other sample types, with the option of contacting Assay Genie technical support for additional matrices.
