Description

Assay Genie offers a Sandwich ELISA Kit (Double Antibody) for the precise measurement of adiponectin levels in porcine samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit employs a colorimetric detection method and is intended for research into adiponectin, described as a key adipokine involved in regulating metabolism and insulin sensitivity, and a relevant biomarker for studying metabolic disorders such as obesity, diabetes, and cardiovascular disease.

Performance figures from the manufacturer include a sensitivity of 37.5 ng/mL and a detection range of 62.5–4000 ng/mL. Linearity data show that for serum (n=5) the values are 93–99% at 1:2, 88–99% at 1:4, and 88–100% at 1:8; for EDTA plasma (n=5) 86–96%, 84–95%, and 83–97% at the same dilutions; and for heparin plasma (n=5) 86–96%, 86–98%, and 88–99%. Recovery averages are 91% for serum (range 86–100%), 93% for EDTA plasma (range 89–100%), and 97% for heparin plasma (range 90–104%).

Storage is recommended at 2–8°C for 12 months. The assay procedure involves equilibrating reagents to room temperature, incubating the plate at 37°C for 90 minutes to allow antigen binding, adding a biotin-labelled detection antibody and incubating for 60 minutes at 37°C, adding HRP-Streptavidin (SABC) and incubating for 30 minutes, developing colour with TMB substrate in the dark for 10–20 minutes, and reading absorbance at 450 nm after adding the stop solution.
