Description

From Assay Genie, this Cell-Based ELISA Kit is designed for the sensitive detection of POLR2A phosphorylated at serine 1619 in cell lysates. It reacts with Human, Mouse, and Rat samples and uses a colorimetric detection method measured at 450 nm, with a dynamic range of > 5000 Cells. The kit is intended for adherent cells and uses an indirect ELISA format, where phospho-specific primary antibodies capture the target and HRP-conjugated secondary antibodies enable detection through a colorimetric reaction upon substrate addition.

The assay is lysate-free and high-throughput, allowing qualitative measurement of relative phosphorylated POLR2A levels in cultured cells. It can be used to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on POLR2A phosphorylation. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalization, while a Crystal Violet whole-cell staining method can be used to determine cell density. POLR2A is the catalytic subunit of RNA polymerase II, and phosphorylation at serine 1619 is known to regulate transcriptional activity and gene expression. Dysregulation of this phosphorylation has been associated with cancer and neurologic disorders, making the kit a useful tool for research into transcriptional regulation, gene expression, and related disease mechanisms. Database identifiers: Gene ID 5430, UniProt ID P24928.
