Description

This Colorimetric Cell-Based ELISA Kit from Assay Genie is designed for detecting POLR2A levels in cell culture supernatants, serum, and plasma samples. POLR2A, also known as RNA polymerase II subunit A, is a vital component of the RNA polymerase II complex, essential for transcriptional activation and gene expression regulation. Dysregulation of POLR2A has been associated with various diseases, including cancer and neurodegenerative disorders, highlighting its relevance as a potential biomarker.

The kit is a convenient, lysate-free, high-throughput and sensitive assay that detects the POLR2A protein expression profile in cells. It is suitable for measuring the relative amounts of POLR2A in cultured cells, including adherent cells, and for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on POLR2A. Qualitative determination of POLR2A concentration is achieved using an indirect ELISA format, in which POLR2A-specific primary antibodies capture the target while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition.

Detection is carried out by a Colorimetric 450 nm method with a dynamic range of > 5000 Cells, and the kit is reactive with Human, Mouse, and Rat samples. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalising target absorbance values. Following colour development, a Crystal Violet whole-cell staining method can be used to determine cell density, so that absorbance values may be normalised to cell amounts. The kit is supplied in a 96-well microplate format, stored at 4°C with a stability of 6 Months. Database references: Gene ID 5430, UniProt ID P24928, OMIM 180660, Unigene Hs.270017.
