Description

From Assay Genie, this Cell-Based ELISA Kit is designed for the precise measurement of phosphorylated PLK1 at serine 137 directly in cultured cells. The assay uses a colorimetric detection method at 450 nm in an indirect ELISA format, offering a lysate-free, high-throughput workflow that detects PLK1 protein phosphorylation and expression without the need for separate lysate preparation. It is reactive with human, mouse, and rat samples, with a dynamic range of greater than 5000 cells, making it suitable for relative quantification studies and for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on PLK1 phosphorylation.

PLK1 (Phospho-Ser137) is captured by target-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary to catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values. Following the colorimetric readout, a Crystal Violet whole-cell staining step may be used to determine cell density, allowing absorbance values to be normalized to cell amounts and adjusting for plating differences.

PLK1, also known as polo-like kinase 1, is a key regulator of cell division and is involved in processes such as mitosis and cytokinesis. Phosphorylation of PLK1 at serine 137 plays a critical role in its activity and function, making this kit a relevant tool for studies of cell cycle regulation and for exploring PLK1 as a potential therapeutic target in cancer and other diseases.
