Description

Assay Genie's Colorimetric Cell-Based ELISA provides a sensitive, lysate-free, high-throughput method for detecting PLCH in cell-based systems, with readout at 450 nm. The indirect ELISA format uses PLCH-specific primary antibodies together with HRP-conjugated secondary antibodies to catalyze a colorimetric reaction upon substrate addition, enabling qualitative measurement of PLCH protein expression in cultured cells and screening of the effects that various treatments, inhibitors, or siRNA have on PLCH levels.

Reactive with human, mouse, and rat samples, the assay is designed for adherent cells and is suited to studying signal transduction pathways, protein trafficking, and protein-protein interactions within cellular systems. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance values, and Crystal Violet whole-cell staining can be used to determine cell density and adjust for plating differences. The dynamic range supports greater than 5000 cells, and results are read from a 96-well microplate format, with storage at 4°C for up to 6 months.

The PLCH target corresponds to the GPAT3 (AGPAT9) enzyme, a 1-acyl-sn-glycerol-3-phosphate acyltransferase involved in glycerolipid biosynthesis and regulation of the TOR signaling pathway.
