Description

From Assay Genie, this Colorimetric Cell-Based ELISA Kit is designed for the sensitive detection of phosphorylated PLCG1 at the Tyr771 residue in cell-based assays. The kit enables researchers to monitor the phosphorylation status of PLCG1 across different cell types and conditions, supporting studies of signal transduction pathways relevant to cancer, autoimmune disorders, and inflammation, and aiding the development of targeted therapies.

The assay uses a convenient, lysate-free, high-throughput indirect ELISA format. PLCG1 (Phospho-Tyr771) is captured by phospho-specific primary antibodies, with HRP-conjugated secondary antibodies enabling colorimetric detection at 450 nm. The kit is reactive with human, mouse, and rat samples and has a stated dynamic range of > 5000 Cells. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance values, and Crystal Violet whole-cell staining may be used to determine cell density, allowing absorbance values to be adjusted for plating differences.

PLCG1 is a calcium-dependent, phosphatidylinositol-specific phospholipase C that generates the second messengers diacylglycerol and inositol 1,4,5-trisphosphate. Phosphorylation at Tyr771, mediated by tyrosine kinases downstream of various growth factor and immune system receptors, regulates PLCG1 activity, making this residue a valuable marker for investigating receptor signalling and for screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on PLCG1 phosphorylation.
