Description

This Colorimetric Cell-Based ELISA Kit from Assay Genie is designed for the accurate detection of phosphorylated PLCG1 (Phospholipase C Gamma 1) at tyrosine 1253 directly in cultured cells. The kit provides a sensitive and specific method for assessing PLCG1 activation status across human, mouse, and rat samples, with a dynamic range of > 5000 cells. It enables researchers to measure the relative amounts of phosphorylated PLCG1 and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on PLCG1 phosphorylation.

The assay uses an indirect ELISA format in which the phosphorylated target is captured by phospho-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. Following substrate addition, HRP catalyses a colorimetric reaction that is read at 450 nm. A monoclonal antibody specific for human GAPDH is included as an internal positive control to normalise target absorbance values, and Crystal Violet whole-cell staining may be used to determine cell density and correct for plating differences.

PLCG1 is a calcium-dependent phosphatidylinositol-specific phospholipase C that produces the second messenger molecules diacylglycerol and inositol 1,4,5-trisphosphate. Phosphorylation at tyrosine 1253, mediated by tyrosine kinases downstream of growth factor and immune receptor signalling, modulates PLCG1 activity and is implicated in cancer, autoimmune disorders, and inflammatory conditions.
