PKR (Phospho-Thr446)Colorimetric Cell-Based ELISA Kit

This colorimetric cell-based ELISA kit from Assay Genie is designed for the detection and qualification of phosphorylated PKR at threonine 446 directly in cultured cells. Using an indirect ELISA format, the assay captures PKR (Phospho-Thr446) with target-specific primary antibodies, while HRP-conjugated secondary antibodies catalyze a colorimetric reaction that is read at 450 nm. The lysate-free workflow supports high-throughput measurement of relative PKR phosphorylation and expression in adherent cell cultures, and is suitable for screening the effects of treatments, inhibitors such as siRNA or chemical compounds, and activators on PKR phosphorylation.

A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalizing target absorbance values, and Crystal Violet whole-cell staining can be used to determine cell density for further normalization to cell amount. Reactivity is with human samples, with a reported dynamic range of greater than 5000 cells. The procedure uses 96-well microplates with overnight primary antibody incubation at 4°C, making the kit a useful tool for research into the role of PKR phosphorylation in innate immune regulation, viral infection, cellular stress responses, autoimmune disorders, and cancer.
