Description

From Assay Genie, this cell-based ELISA kit enables detection of PKD1/PKC mu phosphorylated at serine 205 in cell lysates, with reactivity against human, mouse, and rat samples. The assay uses a colorimetric detection method at 450 nm in an indirect ELISA format, where phospho-specific primary antibodies capture the target and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition.

Designed as a lysate-free, high-throughput assay, the kit measures relative levels of phosphorylated PKD1/PKC mu in cultured adherent cells and can be used to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on PKD1/PKC mu phosphorylation. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density, allowing absorbance values to be normalized to cell amounts and adjusting for plating differences.

The target, PRKD1 (Gene ID: 5587; UniProt ID: Q15139; OMIM: 605435; Gene Symbol: KPCD1), is a CAMK serine/threonine kinase of the PKD family involved in cellular processes including cell proliferation, survival, and differentiation. Phosphorylation at serine 205 is essential for PKD1/PKC mu activation, making this kit a useful tool for investigating signal transduction pathways and protein phosphorylation in research applications.
