Description

From Assay Genie, this In-Cell ELISA Kit provides a lysate-free, high-throughput and sensitive colorimetric assay for detecting PKD1/PKC mu protein expression profiles in cells. The kit is reactive with Human, Mouse and Rat samples and is supplied in a 96-Well Microplate format with detection at 450 nm. It is intended for adherent cell samples and enables relative quantification of PKD1/PKC mu in cultured cells, as well as screening the effects of treatments, inhibitors (such as siRNA or chemicals) or activators on the target.

Qualitative determination is achieved through an indirect ELISA format, in which PKD1/PKC mu-specific primary antibodies capture the target and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalising target absorbance values. A Crystal Violet whole-cell staining method may also be used to determine cell density, allowing absorbance values to be normalised to cell amounts and plating differences to be adjusted. The dynamic range is greater than 5000 cells and the kit is stored at 4°C with 6 months stability.

PKD1 and PKC-Mu are proteins involved in cell signalling pathways that regulate processes such as cell growth, proliferation and differentiation, and their dysregulation has been linked to diseases including cancer, diabetes and autoimmune disorders. Associated database entries include Gene ID 5587, UniProt ID Q15139, OMIM 605435, Unigene Hs.508999 and Gene Symbol KPCD1.
