Description

From Assay Genie, this cell-based ELISA kit is designed for the qualitative detection of Protein Kinase D (PKD) isoforms 1, 2, and 3 in cultured cells. The assay uses a colorimetric detection method read at 450 nm and is provided in a 96-well microplate format with a dynamic range of > 5000 cells. It is validated for reactivity with human, mouse, and rat samples and is intended for use with adherent cells.

This lysate-free, high-throughput assay employs an indirect ELISA format in which PKD1/2/3 is captured by isoform-specific primary antibodies and detected using HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. The kit enables measurement of relative PKD1/2/3 levels in cultured cells and can be used to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), and activators on PKD expression. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance values, and Crystal Violet whole-cell staining may be used to determine cell density for further normalization. The kit is stored at 4°C with a stated stability of 6 months.

PKD proteins are a family of serine/threonine kinases that play roles in cell signaling, regulation of cell proliferation, and modulation of cell migration and invasion. Dysregulation of PKD isoforms has been implicated in various diseases, including cancer, cardiovascular disorders, and inflammatory conditions, making this a useful tool for exploring PKD biology and potential therapeutic interventions.
