Description

From Assay Genie, this Colorimetric Cell-Based ELISA Kit is a lysate-free, high-throughput assay designed to detect Protein Kinase C Beta (PKCβ) protein expression profiles in cultured cells. The assay is intended for use with adherent cells and uses an indirect ELISA approach in which PKCB is captured by PKCB-specific primary antibodies, with HRP-conjugated secondary antibodies binding the Fc region of the primary antibody to catalyze a colorimetric reaction upon substrate addition.

The kit reacts with Human, Mouse, and Rat samples, with a dynamic range of > 5000 Cells and colorimetric detection at 450 nm. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values. Following the colorimetric measurement, the Crystal Violet whole-cell staining method may be used to determine cell density, allowing absorbance values to be normalized to cell amounts and adjusting for plating differences.

Supplied in a 96-Well Microplate format with storage at 4°C for 6 months, the kit can be used for measuring relative amounts of PKCB in cultured cells as well as screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on PKCB. The target is Protein kinase C beta type (NCBI Gene ID: 5579; UniProt ID: P05771), a calcium-activated, phospholipid- and diacylglycerol-dependent serine/threonine-protein kinase involved in multiple cellular processes, including B-cell receptor signaling, oxidative stress-induced apoptosis, androgen receptor-dependent transcription regulation, and insulin signaling.
